Is there a way test this without the Glasses?

Not really, they are only a simple adaptor, not full of parts that could go wrong. Its very unlikely to be faulty, unless its obviously and visibly damaged.

You can grab some cheap generic glasses if don't want to go through the hassle of hunting down an original pair at a reasonable price.

Just wondering does the Mark III have a Card Adapter or is it built in?

The Mark III does have its own seperate adaptor, its white in colour like the system itself and was packaged with the glasses in Japan. The MarkIII adaptor doesn't fit the Master System card slot though.
